Kinetics of impact comminution on a scaled macro-scale

In clinical diagnostics, agricultural science and molecular biological sample preparation, the physical disruption of larger batches of tissue or solid materials (such as bone fragments, infected plant material or seeds) is a process-critical step. The IKA MT 100 sterile disposable grinding jars operate on the process principle of high-speed impact and percussion comminution and extend the working chamber to an isolated 100-ml volume. A solid stainless-steel beater is permanently integrated at the geometric centre of the chamber. Driven by the magnetic coupling of the IKA Tube Mill base station at up to 25,000 revolutions per minute, this rotor shatters macroscopic tissue and cell structures in fractions of a millisecond. The hydrodynamic turbulence (vortex) generated by the expanded volume transforms even larger samples into an analytically homogeneous, fine-powdered matrix that is optimally accessible for subsequent DNA/RNA extraction.

Biological integrity and industrial sterilisation

In PCR-based trace analysis or forensic investigations of representative cross-sectional samples, even the slightest presence of foreign nucleic acids or microbial enzymes (DNases/RNases) leads to a catastrophic metrological false-positive result. The MT 100 sterile beakers are pre-sterilised using a highly controlled industrial process. This physico-chemical process completely breaks down the macromolecular structure of potential biological contaminants inside the grinding chamber. The thermoplastic polymer matrix of the beaker is designed to withstand these sterilisation stresses without any degradation of its structural and ballistic impact resistance, thereby guaranteeing absolute genetic purity (blank value) for the primary analysis buffer in large volumes.

Bioaerosol containment and elimination of carry-over

High-speed grinding of infectious samples or highly potent active pharmaceutical ingredients generates significant fluid dynamic turbulence and internal pressure spikes, leading to the formation of respirable bioaerosols. The MT 100 sterile grinding beaker functions as a hermetically sealed bio-containment system. The process takes place in a completely enclosed atmosphere, thereby physically protecting the user from airborne pathogens. The fundamental design principle as a sterile ‘single-use’ vessel also guarantees that analytical cross-contamination (carry-over) between consecutive patient or environmental samples is completely eliminated, as the error-prone and thermodynamically demanding decontamination of conventional stainless-steel grinding chambers is entirely eliminated.

Individual requirements & accessories

The MT 100.10 sterile grinding jars are highly specialised consumables for large-volume molecular biology or clinical workflows and function exclusively in combination with the IKA Tube Mill drive system. For high-throughput forensic laboratories, pandemic reference centres or in agricultural genetics, where representative sample sections of up to 100 ml need to be processed in a standardised manner, scaling up to large-volume containers (e.g. MT 100.50 sterile) is both economically and logistically essential.
