The Kern OCM 166 is one of the HBO variants in the inverse OCM series, featuring fluorescence filters for UV, V, B and G. A 30-watt halogen lamp provides the brightfield illumination for transmitted light, whilst a 100-watt HBO lamp, which powers the epi-fluorescence unit, provides the reflected light. Kern describes the instrument as an inverted biological laboratory microscope with fluorescence. The head is trinocular; the objectives are Infinity-Plan objectives with 10x, 20x and 40x magnifications, plus a PH 20x phase-contrast objective.

Two light sources for two techniques

The halogen lamp illuminates the specimen from above in brightfield mode and is adjustable. The 100-W HBO lamp from Osram is housed in the epi-fluorescence incident light unit and excites fluorescent specimens from below through the objective. Kern describes the Abbe condenser with an aperture of 0.3 and an aperture diaphragm as suitable for brightfield, phase contrast and fluorescence applications; its working distance is 72 mm. For the halogen light, Kern supplies a 6 V/30 W replacement bulb (OBB-A1372) as an accessory.

Inverted configuration for culture vessels

As the objectives are positioned beneath the stage, the specimen remains accessible from above. Kern highlights the very large working distance of this design for the observation and analysis of cell cultures and describes the design as ergonomic, robust and exceptionally stable. The mechanical stage has a travel range of 128 mm and 180 mm and accommodates a specimen holder for Ø 110 mm. The scope of delivery includes a holder for 54 mm culture dishes; holders for 35 mm and 65 mm dishes are available from Kern as accessories.

Head, eyepieces and focus

The Siedentopf tube is trinocular and inclined at 45 degrees. The HWF 10x eyepieces have a 22 mm field of view, an anti-fungus coating and a high eye point; the interpupillary distance ranges from 48 to 76 mm, and dioptre adjustment is available on both sides. The revolver has five positions; the coaxial coarse and fine adjustment mechanisms operate in increments of 0.002 mm. With the 10x eyepiece, this results in total magnifications of 100x to 400x; with the phase-contrast objective, the magnification is 200x. A centring eyepiece (OBB-A1544) is included in the phase-contrast kit; a 10x eyepiece with a scale is available as an accessory.

Camera and documentation

The third tube port accommodates a camera. Kern supplies C-mount adaptors for this purpose (OBB-A1514, 0.5x with adjustable focus OBB-A1515, 0.75x OBB-A1590) for this purpose, and for fluorescence, the cooled ODC 861 camera, the ODC 825 and ODC 832 microscope cameras, and the ODC 241 tablet camera.

HBO or LED: which OCM

The OCM 167 and OCM 168, on the other hand, feature LED transmitted light and LED reflected light, each with a power of 5 W, and weigh 16 and 17 kg respectively. The OCM 166 weighs 22 kg and has a built-in power supply with a power cord (Protection Class I). If you do not require fluorescence, the OCM 161 will suffice.
