Cell cultures grow in dishes and plates, and anyone wishing to examine them needs space for the culture vessel above the objective lens. The Kern OCM 168 is an inverted microscope designed for this purpose: the objectives are positioned beneath the stage, the specimen rests on top, and, according to Kern, the working distance is very large. Kern describes the instrument as an inverted biological laboratory microscope with fluorescence capability. It features a trinocular head, Infinity-Plan objectives at 10x, 20x and 40x, a PH 20x phase-contrast objective, as well as LED transmitted light and LED reflected light of 5 watts each.
Equipment for culture monitoring
The mechanical stage travels 128 mm in one direction and 180 mm in the other. It is fitted with a specimen holder for Ø 110 mm. A holder for 54 mm culture dishes is included in the Scope of delivery; holders for 35 and 65 mm dishes are available from Kern as accessories. The Abbe condenser has an aperture of 0.3, an aperture diaphragm and a working distance of 72 mm. Kern specifies it for brightfield, phase contrast and fluorescence applications. A phase contrast slider, centring eyepiece and green filter are included as standard.
Lighting
The transmitted light source is a 5 W LED and is dimmable; the incident light source is a second 5 W LED for epi-illumination. Power is supplied via a plug-in power supply unit rated at 100 to 240 V, 50/60 Hz and 0.3 A (Protection Class III); the connection cable is detachable. The OCM 168 is constructed in the same way as the OCM 167, but features a fluorescence filter unit for UV, V, B and G instead of just B and G, and weighs 17 kg instead of 16 kg.
Head and eyepieces
The eyepieces are HWF 10x with a 22 mm field of view, anti-fungal coating and high eye point. The Siedentopf tube is trinocular and inclined at 45 degrees; the interpupillary distance can be adjusted from 48 to 76 mm, and dioptre adjustment is available on both sides. The coaxial coarse and fine adjustment mechanisms have a minimum step of 0.002 mm. With the 10x eyepieces, this results in a total magnification of 100x to 400x. A centring eyepiece is included as standard; an adjustable 10x eyepiece with a scale (0.1 mm) is available as an accessory.
Capturing images
The third tube outlet accommodates a camera. Kern offers a choice of the ODC 854 4K camera (HDMI and USB, Sony sensor), the ODC 251 HD tablet with 4K camera, the ODC 861 cooled camera for fluorescence, the ODC 825 and 832 cameras, and the ODC 241 tablet camera. The corresponding adapters are: C-mount with 0.5x magnification and adjustable focus (OBB-A1515), 0.75x (OBB-A1590) or the standard adapter for trinocular models (OBB-A1514).
Which OCM
The OCM 161 features halogen transmitted light without incident light, whilst the OCM 166 also features a 100 W HBO incident light. Like the OCM 167, the OCM 168 is an LED unit with incident and transmitted light, but features an extended filter set comprising UV, V, B and G.
